Roberta has written an excellent article for the Melungeon project of which she is a DNA advisor. Although, it is written with the Melungeon clan/tribe/group as the focus, however; you can easily apply the same concepts to the Lost Colony Genealogy and DNA Research Group, and others.

Don't skip any of it, thinking I'm not Melungeon, it doesn't apply; the total article discusses points that are applicable, to all of us, and you may miss them. It is written so the person just beginning to understand Genetic Genealogy can fully appreciate the information.
